TOKU-E Announces Ultra-Pure Bio-Acrylamide for Electrophoresis and Other Life Science Applications

BELLINGHAM, Wash.--()--TOKU-E Company (www.toku-e.com) has announced availability of their newly developed, ultra-pure, bioconversion acrylamide.

Two manufacturing processes, chemical synthesis and enzyme conversion, make up current worldwide acrylamide production. Chemical synthesis of acrylamide involves adding water to acrylonitrile, usually catalyzed by the addition of Copper (Cu+). This widely used production method carries three significant disadvantages, including poor yield, unwanted polymerization or conversion to acrylic acid, and difficult regeneration of the Cu+ catalyst.

Tests have shown that enzyme conversion through the use of nitrile hydratase (extracted from Rhodococcus) although used less frequently, yields higher-purity acrylamide. TOKU-E researchers have spent considerable time in optimizing this method as well has downstream purification to increase both yield and purity of the end product. Tests show acrylamide obtained through this production method is 99.9% pure.

Ionic impurities as well as acrylic acid (the main impurity in acrylamide) can have significant effects on the performance of the PAGE gel formed since the voltage across the gel is affected by the ionic charge of the gel and usage buffers. Thus, 99.9% pure acrylamide leads to ultra-low turbidity (<0.5NTU vs. ~2.0 NTU for commercial electrophoresis grade acrylamide) and insoluble impurities of less than 0.005%. This ensures gels of the highest quality, the greatest consistency and ultimately the best resolution.
